    Responses to stimulation of coronary and carotid baroreceptors and the coronary chemoreflex at different ventricular distending pressures in anaesthetised dogs

    Stimulation of left ventricular mechanoreceptors was believed not only to exert important effects on the circulation, but also to influence the responses to baroreceptor reflexes. However, most previous work is flawed due to inadequate localisation of stimuli to specific reflexogenic areas. In this study, we applied a discrete stimulus to left ventricular mechanoreceptors to examine other reflexes known to effect the circulation. Dogs were anaesthetised, artificially ventilated and a cardiopulmonary bypass established. The pressure distending the left ventricle was controlled through an apical cannula with the aortic valve obstructed by a balloon. Changes in ventricular systolic and end-diastolic pressure had only a small effect on vascular resistance, assessed as perfusion pressure in the systemic circulation (flow constant). Responses to changes in carotid or coronary pressure or to stimulation of chemosensitive afferents by injecting veratridine into the coronary circulation were always much larger. Responses to stimulation of these reflexes were little affected by the level of stimulus to the ventricular receptors. These experiments confirm that responses to stimulation of ventricular mechanoreceptors are very small and show that they remain small at different levels of input to other baroreceptive regions. There was no evidence of interaction between ventricular mechanoreceptor reflexes and carotid or coronary baroreceptors or ventricular chemosensitive reflexes

    Evolution of foot-and-mouth disease virus intra-sample sequence diversity during serial transmission in bovine hosts

    RNA virus populations within samples are highly heterogeneous, containing a large number of minority sequence variants which can potentially be transmitted to other susceptible hosts. Consequently, consensus genome sequences provide an incomplete picture of the within- and between-host viral evolutionary dynamics during transmission. Foot-and-mouth disease virus (FMDV) is an RNA virus that can spread from primary sites of replication, via the systemic circulation, to found distinct sites of local infection at epithelial surfaces. Viral evolution in these different tissues occurs independently, each of them potentially providing a source of virus to seed subsequent transmission events. This study employed the Illumina Genome Analyzer platform to sequence 18 FMDV samples collected from a chain of sequentially infected cattle. These data generated snap-shots of the evolving viral population structures within different animals and tissues. Analyses of the mutation spectra revealed polymorphisms at frequencies >0.5% at between 21 and 146 sites across the genome for these samples, while 13 sites acquired mutations in excess of consensus frequency (50%). Analysis of polymorphism frequency revealed that a number of minority variants were transmitted during host-to-host infection events, while the size of the intra-host founder populations appeared to be smaller. These data indicate that viral population complexity is influenced by small intra-host bottlenecks and relatively large inter-host bottlenecks. The dynamics of minority variants are consistent with the actions of genetic drift rather than strong selection. These results provide novel insights into the evolution of FMDV that can be applied to reconstruct both intra- and inter-host transmission routes

    Role of COX-1 and COX-2 in the release of prostanoids in murine lung and isolated lung fibroblasts

    Cyclooxygenase (COX) is the first enzyme in the conversion of arachidonic acid to prostanoids. There are two isoforms of COX; COX-1, which is constitutively expressed with a homeostatic role in most tissues, and COX-2, which while constitutively expressed in some discreet sites is generally inducible by growth factors and during inflammation. In the current study, we have used tissues and cells from knock-out mice to investigate the relative contributions of COX-1 and COX-2 to PGE2 production by lung tissue ex vivo and by proliferating lung fibroblasts in vitro. Lung tissues from WT (C57Bl6), COX-1-/- and COX-2-/- mice were immediately dissected (<15 min after death) and incubated (37 °C) for 30 min in DMEM containing 50 µM calcium ionophore (A23187). Release of PGE2 was determined by competitive immunoassay. In parallel studies, murine lung fibroblasts from COX-1-/- and COX-2-/- mice were explanted and cultured before being seeded in 96-well plates at sub-confluence (5000-8000/well) and incubated for 24-48 hours in the presence of 10% FCS. Accumulated release of PGE2 was then measured as above. Over 30 min PGE2 was released by lung pieces from wild type (1117 ± 55 pg/ml) and COX-2-/- (2013 ± 255 pg/ml) but not from COX-1-/- (<61pg/ml) mice (n=4). In contrast, proliferating lung fibroblasts from COX-1-/- (4978.9 ± 1392 pg/ml) mice released higher levels of PGE2 than cells from COX-2-/- (1194 ± 617 ng/ml) mice (n=4 using cells from 2-3 separate mice for each genotype). These results show that COX-1 activity underpins the stimulated release of PGE2 in healthy mouse lung tissue. Conversely, COX-2 activity predominates in proliferating lung fibroblasts, which may be important as COX-derived PGE2 mediates proliferation of lung fibroblasts (Trends Immunol.2004;25(1):40-6). Our results suggest a switch in COX isoform in lung cells during proliferation which could be relevant to our understanding of conditions such as idiopathic pulmonary fibrosis.Non peer reviewedFinal Accepted Versio

    Student perceptions and experiences of problem-based learning in first year undergraduate sports therapy

    Problem-based learning (PBL) has long been used as a means to foster critical thinking and student autonomy. However, few studies have investigated the effectiveness of PBL in Sports Therapy. The aim of this study was to examine first year Sports Therapy students' perceptions of PBL. Results revealed that students perceived PBL as vocationally relevant, by enabling them to work in and across groups, whilst also engaging with a wider range and depth of information compared to more traditional methods of curriculum delivery. External observations of the lecturers input sessions were made by an impartial re- searcher. The implications of the study are that PBL appears to be a professionally suitable and appropriate learning modality for Sports Therapy students

    Feshbach Resonances and Medium Effects in ultracold atomic Gases

    We develop an effective low energy theory for multi-channel scattering of cold atomic alkali atoms with particular focus on Feshbach resonances. The scattering matrix is expressed in terms of observables only and the theory allows for the inclusion of many-body effects both in the open and in the closed channels. We then consider the frequency and damping of collective modes for Fermi gases and demonstrate how medium effects significantly increase the scattering rate determining the nature of the modes.
